金焰彩欒組織培養技術研究

摘要:通過正交試驗等方法開展的金焰彩欒離體培養技術研究結果表明:從金焰彩欒播種后生長3個月的苗上選取外植體,經過0.1% HgCl2消毒5 min,滅菌成活率為86.8%;改良MS培養基附加BA,NAA,IBA、IAA等植物生長調節劑可有效促進試管芽苗增殖和生根,其中BA 0.02 mg/L+IBA 0.02 mg/L+NAA 0.03 mg/L+IAA 0.08 mg/L組合培養基為最適宜的繼代培養基,增殖倍數達3.8,生長高度為4.8 cm;帶根試管苗移栽于泥炭(V)∶黃心土(V) =6∶4的混合基質,控制溫度23~30℃,相對濕度85%,保濕10~15 d,其間適當遮蔭,20 d后成活率達92.8%。

The in vitro tissue culture of Koelreuteria bipinnata var integrifoliola‘Jinyancailuan’

Abstract:We studied the technology of in vitro tissue culture of Koelreuteria bipinnata var integrifoliola‘Jinyancailuan’by orthogonal test.The results showed that after disinfection with 0.1% HgCl2for 5 min,the survival rate of the explants excised from three-month old seedlings could reach 86.8%.The modified MS media supplemented with BA,NAA,IBA or IAA could elevate effectively the tube shoot proliferation and rooting rate,among which,the medium containing 0.02 mg/L BA+0.02 mg/L IBA+0.03 mg/L NAA+0.08 mg/L IAA was most suitable for subculture with proliferation rate of 3.8 and inoculated plantlets height of 4.8 cm.After transferred into the mixed substrate composed of peat and yellow soil (6∶4,V/V),the rooted plantlets could gain the survival of 92.8% 20 d later in case of temperature controlled at 23~30℃,relative humidity of 85%,moisturizing lasted for 10~15 d with proper shading.

黃山欒樹,為無患子科(Sapindaceae)欒樹屬(Koelreuteria)樹種,枝葉茂密,樹形優美。春季,紅葉引人矚目,夏秋黃葉耀眼,秋果紫紅,酷似燈籠,奇特美麗,具有較強的綠化、觀賞價值[1-2]。前人在其播種繁殖、芽苗移栽技術[3-7]、種子脂肪分析、果實維生素含量、葉清除自由基活性物質的分離和制備、體細胞胚的發生和組織學觀察等方面開展了研究[8-12]。金焰彩欒(Koelreuteria bipinnata var integrifoliola‘Jinyancailuan’)是在黃山欒樹中選出的優良品種,其主要特性是樹干或枝條呈黃色,春季新芽、葉呈桔紅色,夏季葉呈黃綠色,秋葉呈金黃色,具有很高的園藝觀賞價值[13]。在現有技術中,主要采用嫁接方法進行繁殖,尚未有組織培養技術研究的報道[13]。本文對金焰彩欒組織培養技術進行了探索,試圖加快規模化繁育速度,滿足園林綠化市場和林業生態建設的需求。
