牛蒡子化學成分的研究

牛蒡子為菊科植物牛蒡(Arctium lappaL.)的干燥成熟果實,屬辛涼解表藥具疏散風熱、宣肺透疹、消腫解毒之功效。用于治療風熱感冒、咳痰多、咽喉腫痛、斑疹不透、風疹作癢、癰腫瘡毒等癥,在我國分布廣泛[1]。其脂肪油營養不亞于核桃油、大豆油。牛蒡葉多作外用,有明顯的消炎、解毒、鎮痛作用。牛蒡根,又名東洋參,有明顯的降血糖、降血脂、降血壓、補腎壯陽、潤腸通便和抑制癌細胞滋生、擴散及移棄水中重金屬的作用,是非常理想的天然保健食品,在國內外市場上作為蔬菜均有銷售,享有“蔬菜之王”的美譽[2,3]。為了解其里面的主要化學成分,我們對買自昆明菊花村藥材市場的牛蒡子進行化學成分研究。通過柱層析,制備HPLC 和現代波譜學技術分離鑒定了14 個木脂素類化合物,分別為牛蒡子苷(1),牛蒡苷元(2),羅漢松樹脂酚(3),牛蒡酚B(4),異牛蒡酚A(5),牛蒡酚A(6),(+)-Diasyringaresinol(7),tanegool(8),arctignan F(9),牛蒡酚F(10),牛蒡酚C(11),arctignan D(12),arctignan E(13),牛蒡酚H(14),其中化合物7 和8 為首次從該種植物中分離得到。

1 儀器與試劑

1D-NMR 在Bruker AM-400、Bruker DRX-500或Bruker AM-600 核磁共振儀上測定,TMS 作為內標,δ 為ppm,J為Hz;ESI-MS 在Burker HCT 或Esquire 質譜儀上測定;高分辨質譜在Auto-Spec Premier P776 質譜儀上測定;拌樣及層析用硅膠(100-200,200~300 目),均為青島海洋化工廠生產;反相填充材料RP-18 為40~60 μm,Merk 公司生產;MCI填充材料為MCI-gel CHP-20P;HPLC 分析儀器為Agilent 1260 型高效液相色譜儀,色譜柱為Agilent公司的ZORBAX SB-C18反相柱(9.6 ×150 mm,5 μm)。凝膠為Sephadex LH-20;顯色劑非堿為10%H2SO4的乙醇溶液,噴灑后適當加熱。

實驗藥材購自于云南昆明菊花村藥材市場。由中國科學院昆明植物研究所植物園成曉研究員鑒定。

2 提取與分離

干燥牛蒡子21 kg,粉碎后用甲醇提取三次(50 L/次),合并提取液,減壓蒸餾除去有機溶劑,得到粗提物1.4 kg,將此粗提物用2.0 kg 100~200 目硅膠拌樣,以不同比例的石油醚-丙酮系統(9∶1,8∶2,7∶3,1∶1,0∶1)進行梯度洗脫,用TLC 進行檢測,合并相同組分得到七個餾分:I-Ⅶ。

餾分Ⅳ用丙酮結晶,得到化合物1(35.4 g)。

餾分Ⅶ以聚酰胺拌樣,以甲醇-水系統(45%-100%)在MCI 柱上進行梯度洗脫,以TLC 進行檢測,合并相同組分得到Ⅶa,Ⅶb,Ⅶc,Ⅶe,Ⅶf 和Ⅶg。Ⅶa 段先以不同比例的氯仿-丙酮系統(95∶5-1∶1)進行梯度洗脫,用TLC 進行檢測,合并相同組分,然后再以甲醇-水系統(23∶87,流速3 mL/min)進行半制備,得到化合物3 (181.8 mg,tR=12.8 min),4(1.5 g,tR=12.9 min),和5 (42.2 mg,tR=19.3 min)。Ⅶb 段經過多次柱層析得到化合物6(6.1 mg)和7(2.8 mg)。Ⅶc 用甲醇結晶,得到化合物2(55.89 g),對其渾濁液以甲醇-水系統(43∶57,流速3 mL/min)進行半制備,得到化合物10(121.0 mg,tR=15.2 min)。Ⅶd 段經過多次柱層析得到化合物8(57.5 mg)和11(54.2 mg)。Ⅶf 段以乙腈-水系統(23∶77,流速3 mL/min)進行半制備,得到化合物12 (55.1 mg)和13 (70.0 mg)。Ⅶg 段以甲醇-水系統(45∶55,流速3 mL/min)進行半制備,得到化合我9(4.4 mg,tR=13.6 min)和14(26.9 mg,tR=19.3 min)。

3 結構鑒定
