摘要 將一株高產酸植物乳桿菌,在含有馬鈴薯秧藤汁液的一組MRS液體培養基中連續轉移接種培養,組內5個培養基中的秧藤汁液濃度呈階梯性遞增狀態,對完成連續馴化培養后的菌種進行冷凍干燥保存。進一步將該馴化菌和未馴化菌進行同步解凍激活后,分別接入含有馬鈴薯秧藤固體顆粒的液體培養基中培養48 h,結束后分離出各自培養基中的馬鈴薯秧藤固體顆粒部分和液體部分,采用比色法分別測定固體和液體部分中糖苷生物堿的含量;分別測定液體部分的pH值和總細菌細胞數。結果表明:在馴化菌的培養體系中,總細菌細胞數含量顯著高于對照組;pH值顯著低于對照組;馬鈴薯秧藤固體顆粒中糖苷生物堿含量明顯低于對照組;培養液中的糖苷生物堿顯著高于對照組。

Study on the Cultivation of Lactobacillus of Adaptive Acclimation to the Juice of Potato Cane

Abstract In the experiment,the plant lactobacillus which have high acid production was transferred into a group of MRS liquid nutrient medium for the continuous transfer and cultivation,In this group of 5 liquid medium the juice of potato cane was the ladder of increasing state.Then,the freeze drying of the strains after continuous cultivation was carried out.Further,after thawing activation of the domesticated bacteria and not domesticated bacteria synchronously,They were inoculated to the liquid nutrient medium which included the juice of potato cane cultivate 48 h.After the training the potato cane solid particles and liquid parts in the medium was each separated.The contents of the Glycosides alkaloids in the solid and liquid parts were determined respectively by colorimetric method;pH values and total bacterial count were determined in the liquid parts.The results showed that in the cultivation system of the domesticated bacteria,the total bacterial cell number was significantly higher than that of the control group;pH value was significantly lower than that of the control group;Glycosides alkaloids content in the potato cane solid particles was significantly lower than that of the control group;Glycosides alkaloids in the liquid were significantly higher than those in the control group.

馬鈴薯秧藤是馬鈴薯植株的地上部分,是在收獲塊莖后剩余的副產品。在傳統的馬鈴薯種植業中,秧藤一般作為廢棄物被處理;在現代化的馬鈴薯種植業中,為促進塊莖薯皮提早老化、便于機械收獲土豆作業以及預防各類病原體傳播,一般在土豆收獲前幾天至十幾天,采用化學殺秧、機械打秧等方式,將秧藤打碎還田或清除出田地的田間管理措施,這使得馬鈴薯秧藤資源具有了容易收集、分布集中、數量龐大的可開發利用優勢。研究表明,馬鈴……
