摘要:目的 檢測乳腺癌患者mtDNA D-loop環(huán)HV2區(qū)體細(xì)胞性突變并探討它在腫瘤發(fā)生中的作用。方法 采用聚合酶鏈反應(yīng)-單鏈構(gòu)像多態(tài)性(PCR-SSCP)的方法對20例浸潤性乳腺導(dǎo)管癌組織和癌旁正常組織的mtDNA非編碼區(qū)D-loop環(huán)HV2區(qū)進(jìn)行分析,將出現(xiàn)異常條帶所有標(biāo)本進(jìn)行基因測序。結(jié)果 經(jīng)PCR-SSCP檢測,在20例乳腺癌mtDNA中共發(fā)現(xiàn)7例出現(xiàn)異常條帶;測序結(jié)果顯示,7例測序標(biāo)本中,HV2區(qū)共檢測到31個(gè)新的突變位點(diǎn),其中有3個(gè)屬于微衛(wèi)星不穩(wěn)定性(microsatellite instability,MSI),7例移碼突變,其余為點(diǎn)突變(point mutation),7個(gè)位點(diǎn)位于復(fù)制和轉(zhuǎn)錄的區(qū)域。結(jié)論 浸潤性乳腺導(dǎo)管癌mtDNA HV2區(qū)是一個(gè)高度多態(tài)性和突變性的區(qū)域,它與乳腺癌的發(fā)生、發(fā)展有重要關(guān)系。

Abstract:Objective To detect the effect of mutations of HV2 region of D-loop in mtDNA of the breast cancer.Methods It analysed the HV2 region of D-loop in mtDNA in the 20 cases invasive ductual breast carcinoma and 20 cases paracarcinoma tissues by PCR and PCR-single stranded conformation polymorism.Results The 7 cases exceptional bands were found from 20 breast carcinoma in all of the samples by PCR-SSCP;and the sequencing results indicated that 31 new HV2 mutations were found in 7 sequencing specimens,the 3 of them were mitochondrial microsatellite instability(MSI);the 7 of them were frameshift mutations;others were point mutation;and the 7 of them located in the region of mtDNA replication and transcription.Conclusion The mtDNA HV2 region is a highly polymorphic and mutatble region,that could be some relations in the breast carcinoma development.

線粒體DNA(mitochondrial DNA,mtDNA)存在于細(xì)胞質(zhì)的線粒體中,是細(xì)胞內(nèi)唯一的核外遺傳物質(zhì),由16569bp組成的雙鏈閉合環(huán)狀分子,含有自我復(fù)制、轉(zhuǎn)錄和翻譯的遺傳體系。mtDNA含有37個(gè)編碼基因,包括編碼氧化磷酸化系統(tǒng)13個(gè)蛋白質(zhì)的亞單位、2個(gè)rRNA及22個(gè)tRNA的結(jié)構(gòu)基因。由于不受組蛋白保護(hù),修復(fù)功能不完善,很容易成為致癌物質(zhì)攻擊的標(biāo)靶。因此mtDNA的突變與細(xì)胞癌變之間的關(guān)系已經(jīng)越來越受到重視,成為研究的熱點(diǎn)。本文利用PCR-SSCP和基因測序法對mtDNA D-loop環(huán)HV2區(qū)的基因突變情況進(jìn)行了研究,并探討了突變在乳腺癌進(jìn)程中的作用。
1 資料與方法
1.1一般資料 取自2004年5月~2005年7月延邊大學(xué)附屬醫(yī)院腫瘤外科手術(shù)的患者,20例乳腺癌組織及相對應(yīng)的癌旁正常組織,手術(shù)后切除部分組織立即投入液氮中,后放入-80℃冰箱中保存?zhèn)溆谩!?br>
