摘 要:該研究為彌漫性腦損害(精神分裂癥)的腦網絡研究(2011CB707805)一般性科技報告,報告概括了研究思路、技術路線、實驗方案并總結2011—2013度計劃執行情況;年度計劃完成情況;研究工作的主要進展;重要階段性成果或突破;項目執行過程中存在的問題及其對策及下年度研究工作計劃和進度安排對年度情況進行匯總。該項目圍繞研究目標,依照研究方案針對患者、高危人群與正常對照,分別研究腦結構網絡和功能網絡,分析各自的網絡屬性,找尋疾病所產生的異常模式。利用多中心數據發現了:(1)腦結構網絡異常:精神分裂癥患者皮層厚度、灰質體積及白質解構的異常,病分析高危人群:精神分裂癥患者及其父母大腦杏仁核存在共同的結構和功能連接異常模式;(2)腦功能網絡研究:精神分裂癥的腦解剖網路研究 精神分裂癥病人和正常對照被試的的腦解剖網絡均具有小世界屬性,但是精神分裂癥患者腦網絡的全局連接效率降低,精神分裂癥患者不同臨床和認知功能評定指標與各種腦網絡指標的相關性的研究;(3)腦網絡相關生物基因標記物關聯研究:精神分裂癥風險基因與腦網絡的研究表明DISC1不同基因型對于丘腦-皮層通路的結構與功能連接有著顯著不同的影響,DISC1基因的Cys等位基因攜帶者腦網絡信息傳輸效率較低,5-HTTLPR與情緒相關的關系以及對杏仁核-前額環路的影響。從影像學及生物學標記物層面對腦網絡進行了深入研究。揭示彌漫性腦損害(精神分裂癥)對腦結構和功能網絡影響的本質特性,在網絡水平、基因水平上發現精神分裂癥早期診斷、預后判斷及療效評價的影像學標志,生物學標記為這些疾病的臨床早期診斷和療效評價等提供有效的方法和途徑。

Abstract:This is the scientific summary of Brainnetome research in patients with diffuse lesions(schizophrenia, No.2011CB707805),which summarizes the studies designs, techniques, protocols and summarizes research results from the 2011-2013. According to the multi- center data, abnormal brain structure, functional network, and the network property's patterns of the disease in the schizophrenia patients group, high-risk group and normal controls were discovered:(1)the structural abnormalities of the brain network were discovered in patients with schizophrenia,such as the cortical thickness , the gray matter volume and the white matter deconstruct. The patients with schizophrenia and their parents presence abnormal of amygdala functional connectivity ;(2)Functional brain network studies: the patients with schizophrenia brain presence small world properties, anatomical networks study between the patients with schizophrenia, and the normal control subjects showed that lower global brain network connective efficiency in patients with schizophrenia,which correlated with different clinical and cognitive function in patients with schizophrenia. (3)Schizophrenia risk genes and brain networks studies have shown that DISC1 different genotypes for the hypothalamus cortical connections has different effects, DISC1 risk genes significantly lower Cys allele brain network efficiency. From the MR imaging and biological markers of brain network level, we revealed the characteristics in brain structural and functional network effects in diffuse brain lesions ( patients with schizophrenia ). The research results may become the radiographic bio-marker in schizophrenia's early diagnosis, prognosis and treatment evaluation on the network level.
