摘要[目的]運用生物信息學方法預測兩歧雙歧桿菌PRL2010錨定于細胞壁的蛋白和功能分析,為后續研究該菌與宿主相互作用的分子機制奠定基礎。[方法]用Phobius和SignalP 4.0軟件對兩歧雙歧桿菌PRL2010全基因組編碼蛋白中細胞壁蛋白進行預測,同時采用COG(Cluster of Orthologous Groups of proteins)功能數據庫對預測的細胞壁蛋白進行功能注釋和聚類分析。[結果]兩歧雙歧桿菌PRL2010基因組1 706個編碼蛋白中,28個蛋白含有細胞壁的錨定結構域。細胞壁蛋白功能分析結果顯示,28個細胞壁蛋白中,21個蛋白沒有功能注釋,7個蛋白有功能注釋,其中1個蛋白(exoalphasialidase)參與碳水化合物代謝與轉運,2個蛋白(bacteriophage endolysin和cell wallassociated protein)參與細胞壁和細胞膜的生物合成,2個蛋白(hypothetical protein BBPR_0440和Subtilisin family peptidase)參與蛋白質翻譯后修飾,1個蛋白(betanacetylhexosaminidase NagZ)參與細胞內運輸、分泌和囊泡運輸,1個蛋白(metallophosphoesterase)功能只是預測的。[結論]兩歧雙歧桿菌PRL2010的大多數細胞壁蛋白功能未知,還需要在以后的研究中進一步分析和注釋。

Prediction and Functional Analysis of Cell Wall Proteins of Bifidobacterium bifidum PRL2010

Abstract[Objective]The genome sequences of Bifidobacterium bifidum PRL2010 were used to predict the cell wall proteins and functional analysis, which will provide the basis for molecular mechanisms of interaction between bacteria with their host. [Method] Phobius and SignalP 4.0 softwares were used to analyze the cell wall proteins. Function of cell wall proteins was also analyzed by COG (Cluster of Orthologous Groups of proteins) database. [Result] There were 28 cell wall proteins which possessed cell wall motif from 1 706 proteins coded by B. bifidum PRL2010 genome.Function analysis revealed that the strain contains 21 cell wall proteins without function annonation.There were 7 proteins that having function annonation which involved in metabolism and transport of carbohydrate(1 protein: exoalphasialidase), cell wall/membrane biosynthesis (2 proteins: bacteriophage endolysin and cell wallassociated protein), posttranslational modification (2 proteins: hypothetical protein BBPR_0440 and Subtilisin family peptidase), intracellular trafficking,secretion and vesicular transport(1 protein: betanacetylhexosaminidase NagZ),function prediction(1 protein: metallophosphoesterase). [Conclusion] Function of most cell wall proteins of B. bifidum PRL2010 was unknown, which need to be researched and annotated in the future research.

兩岐雙歧桿菌PRL2010最早分離于嬰兒糞便中,具有抑制腸道致病菌黏附[1],降低血清膽固醇[2],調節宿主免疫[3],調整腸道菌群[2]等益生作用,能夠降解宿主衍生的黏蛋白[3-4]的獨特特征。兩岐雙歧桿菌PRL2010作為一種革蘭氏陽性菌,菌體最外含有一層較厚的細胞壁,主要是由肽聚糖和磷壁酸組成。在細胞壁的外周有時錨定有蛋白質,這些蛋白質通過特殊的結構錨定在細胞壁上,如含有LPxTG結構的區段、LysM、PG等區域,由于細胞壁蛋白處于菌體的外層,在適應外界環境和與宿主細胞之間發生相互作用方面,具有重要的作用。……
