野木瓜莖枝三萜皂苷類化學成分研究△

野木瓜StauntoniachinensisDC.系木通科Lardizabalaceae野木瓜屬植物,是廣泛分布在我國長江以南地區的野生藥用植物,全株具有較高的藥用價值[1]。野木瓜味微苦,性平,歸肝、胃經,具有祛風止痛、舒筋活絡的功能。中醫常用于風濕痹痛、腰腿疼痛、頭痛、牙痛、痛經、跌打傷痛[2]。野木瓜經提取加工制成的制劑有野木瓜注射液、野木瓜片等,已廣泛應用于臨床。現代醫學研究表明野木瓜主要的藥理作用是鎮痛抗炎、阻滯神經傳導和放射增敏等。臨床上主要用于術后傷口痛、風濕痛、各種神經性疼痛、內臟疼痛、腰椎骨質增生等,療效顯著,且具有無耐藥性、無成癮性、不良反應小等特點[3]。目前,國內外學者報道了野木瓜中不僅含黃酮苷、酚酸、糖等類成分,還含有豐富的皂苷[4],主要包括五環三萜皂苷類化合物,其具有多種生物活性,如護肝[5]、抑癌[6-7]、抗炎[8]、抗氧化[9],以及降血糖和增強免疫能力[10]等藥理作用。本文重點對野木瓜中皂苷類成分進行了研究,利用各種色譜技術從野木瓜莖枝水提物的正丁醇部位中分離鑒定了11個三萜皂苷類化合物,分別為:野木瓜苷YM30(1),野木瓜苷YM23(2),野木瓜苷YM12(3),野木瓜苷YM14(4),野木瓜苷YM25(5),3-O-α-L-吡喃鼠李糖基-(1→2)-α-L-吡喃阿拉伯糖基-30-降常春藤皂苷元-28-O-α-L-吡喃鼠李糖基-(1→4)-β-D-吡喃葡萄糖基-(1→6)-β-D-吡喃葡萄糖基酯苷(6),野木瓜苷YM8(7),3β,20α,24-三羥基-29-降齊墩果-12-烯-28-羧酸-23-O-β-D-吡喃葡萄糖苷(8),3β,20α,24-三羥基-29-降齊墩果-12-烯-28-羧酸-23-O-β-D-吡喃海藻糖基-(1→2)-[α-L-吡喃阿拉伯糖基-(1→4)]-β-D-吡喃葡萄糖苷(9),3-O-β-D-吡喃木糖基齊墩果酸-28-O-α-L-吡喃鼠李糖基-(1→4)-β-D-吡喃葡萄糖基-(1→6)-β-D-吡喃葡萄糖基酯苷(10),3-O-β-D-吡喃葡萄糖基-(1→4)-β-D-吡喃木糖基-(1→3)-α-L-吡喃鼠李糖基-(1→2)-α-L-吡喃阿拉伯糖基齊墩果酸-28-O-β-D-吡喃葡萄糖基-(1→6)-β-D-吡喃葡萄糖苷(11)。化合物8~11為首次從野木瓜中分離得到。

1 儀器與材料

Buchi中壓液相制備色譜儀(瑞士步琪公司);島津2010系列高效液相色譜儀(日本島津公司);安捷倫1200型制備高效液相色譜儀;Sartorius BP211D型電子天平(德國賽托利斯集團);UV-260分光光度計(日本島津公司);Micromass ZabSpec質譜儀(美國Micromass公司);Bruker DRX-400超導核磁共振儀(德國Bruker 公司);C18反相填料為日本YMC產品;Sephadex LH-20為瑞典Amersham Biosciences生產;柱色譜和薄層色譜硅膠均為青島海洋化工廠生產;水為雙蒸水;氘代試劑C5D5N(美國劍橋公司CIL);其他所用試劑均為分析純。

野木瓜采自江西贛州,經江西中醫藥大學劉勇教授鑒定為木通科野木瓜屬植物野木瓜StauntoniachinensisDC.干燥莖枝。標本保存在江西省藥品檢驗檢測研究院中藥標本室。

2 提取與分離

野木瓜干燥莖枝40 kg,粉碎,用6倍量的水加熱回流提取3次,每次2 h,提取液濃縮至密度為1.15,依次使用三氯甲烷、乙酸乙酯、正丁醇萃取。萃取液分別濃縮至干膏,得到三氯甲烷萃取部分(50 g)、乙酸乙酯萃取部分(105 g)和正丁醇萃取部分(550 g)。取正丁醇部分干膏100 g,用1∶1.5的硅膠拌樣,經正相硅膠柱色譜,以二氯甲烷-甲醇(2∶1)為洗脫劑,切干柱得到10個組分Fr.1~Fr.10。Fr.2(4.5 g)以三氯甲烷-甲醇(6∶1~1∶1)梯度洗脫,經薄層色譜和高效液相色譜檢測合并相似餾分得10個組分Fr.2-1~Fr.2-10。Fr.2-4(2.2 g)經中壓制備液相色譜,以甲醇-水(20∶70~90∶10)梯度洗脫,經高效液相色譜檢測,合并相近組分,得到4個組分Fr.2-4-1~Fr.2-4-4,其中Fr.2-4-2(161 mg)經反相高效制備液相色譜,以乙腈-水(22∶78)(7 mL·min-1)為流動相,得到化合物8(3.4 mg)。Fr.2-4-3(68 mg)經反相高效制備液相色譜,以乙腈-水(22∶78)(7 mL·min-1)為流動相,得到化合物9(7.2 mg)。Fr.7(10 g)經中壓制備液相色譜,以二氯甲烷-甲醇(10∶1~1∶1)梯度洗脫,經薄層色譜和高效液相色譜檢測合并相似流分,得到10個組分Fr.7-1~Fr.7-10。Fr.7-3(1.5 g)經中壓制備液相色譜,以甲醇-水(30∶70~90∶10)為流動相進行梯度洗脫,經高效液相色譜檢測合并相近的組分,得到5個部分Fr.7-3-1~Fr.7-3-5,其中Fr.7-3-2(380 mg)經反相高效制備液相色譜,以乙腈-水(28∶72)(7 mL·min-1)為流動相,得到化合物1(4.2 mg)、2(4.7 mg)和3(7.2 mg)。Fr.7-3-5(130 mg)經反相高效制備液相色譜,以乙腈-水(26∶74)(7 mL·min-1)為流動相,得到化合物4(3.5 mg)。Fr.8(6.3 g)以二氯甲烷-甲醇(12∶1~1∶1)為流動相進行梯度洗脫,經薄層和高效液相色譜檢測,合并相近流分,得到10個組分Fr.8-1~Fr.8-10。Fr.8-4(2.0 g)經中壓制備液相色譜,以甲醇-水(20∶70~90∶10)梯度洗脫,經高效液相色譜檢測,合并相近流分,得到5個部分Fr.8-4-1~Fr.8-4-5,其中Fr.8-4-3(420 mg)經反相高效制備液相色譜,以乙腈-水(30∶70)(7 mL·min-1)為流動相,得到化合物5(12.2 mg)和6(6.2 mg)。Fr.9(4.2 g)經正相硅膠柱色譜,以二氯甲烷-甲醇(13∶1~1∶1)為流動相進行洗脫,經高效液相色譜檢測,合并所得流分中相近的組分,得到10個組分Fr.9-1~Fr.9-10。Fr.9-3(1.8 g)經中壓制備液相色譜,以甲醇-水(20∶70~90∶10)梯度洗脫,經高效液相色譜檢測,合并所得流分中相近的部分,得到5個部分Fr.9-3-1~Fr.9-3-5,其中Fr.9-3-3(1.5 g)經反相高效制備液相色譜,以乙腈-水(32∶668)(7 mL·min-1)為流動相,得到化合物7(3.1 mg)。Fr.10(1.8 g)經正相硅膠柱色譜,以二氯甲烷-甲醇(7∶1~1∶1)為流動相進行梯度洗脫,經薄層色譜檢測,合并所得流分中相近的部分,得到8個組分Fr.10-1~Fr.10-8。Fr.10-5(600 mg)經中壓制備液相色譜,以甲醇-水(20∶70~90∶10)梯度洗脫,經高效液相色譜檢測,合并所得流分中相近部分,得到4個部分Fr.10-5-1~Fr.10-5-4,其中Fr.10-5-4(60 mg)經反相高效制備液相色譜,以乙腈-水(28∶72)(7 mL·min-1)為流動相,得到化合物10(4.6 mg)和11(5.7 mg)。

3 結構鑒定
