LncRNA與炎癥性疾病關(guān)系的研究進展*

長鏈非編碼RNA(lncRNA)是一類缺少完整開放閱讀框(ORF)的非編碼RNA,在細胞增殖、分化和凋亡等生物學(xué)過程中發(fā)揮重要作用。目前研究發(fā)現(xiàn),lncRNA在炎癥反應(yīng)和炎癥性疾病中表達異常,可通過調(diào)節(jié)多種基因表達和信號通路激活,參與炎癥性疾病的發(fā)生、發(fā)展。本文就lncRNA與炎癥性疾病關(guān)系的研究進展作一綜述。

AbstractLong non-coding RNA (lncRNA) is a non-coding RNA which lacks a complete open reading frame (ORF) and plays an important role in biological processes such as cell proliferation, differentiation and apoptosis. At present, expression of lncRNA has been found to be abnormal in inflammatory reaction and inflammatory diseases. It might be involved in the development and progression of inflammatory diseases via regulating the expression of multiple genes and activation of signaling pathways. This article reviewed the progress in study on relationship between lncRNA and inflammatory diseases.

炎癥是由多種炎癥細胞和炎癥因子共同參與的重要病理過程,是機體對病原微生物感染、創(chuàng)傷、變態(tài)反應(yīng)等發(fā)生的組織細胞反應(yīng)。長鏈非編碼RNA(lncRNA)是一類缺少完整開放閱讀框(ORF)的非編碼RNA,對基因表達、細胞分化、細胞凋亡乃至疾病(如炎癥、腫瘤)均有重要影響,在基因轉(zhuǎn)錄和轉(zhuǎn)錄后調(diào)控等過程中扮演重要角色,如X染色體失活、基因組印記、干細胞多能性、癌癥轉(zhuǎn)移、動脈粥樣硬化等[1]。lncRNA在炎癥性疾病中異常表達,與多種炎癥相關(guān)基因表達和信號通路活化有關(guān)。lncRNA對炎癥反應(yīng)的調(diào)節(jié)作用為炎癥性疾病的診治提供了新思路。本文就lncRNA與炎癥性疾病關(guān)系的研究進展作一綜述。
lncRNA是一類長度大于200 nt的非編碼RNA,因缺少完整的ORF而無蛋白質(zhì)編碼功能。據(jù)報道,lncRNA的轉(zhuǎn)錄與mRNA類似,多數(shù)由RNA聚合酶Ⅱ介導(dǎo),且通常包含多聚腺苷酸化信號序列;部分lncRNA轉(zhuǎn)錄可由RNA聚合酶Ⅲ介導(dǎo)[2]。相比蛋白質(zhì)編碼基因,大多數(shù)lncRNA的保守性低于mRNA,但并不一定意味其缺乏功能。……
