摘要 研究小黑藥全株總黃酮的含量及提取分離,并用Schaal烘箱法、紫外光照法測(cè)試了小黑藥總黃酮和對(duì)照品BHT抗豬油氧化的能力,采用POV抑制率評(píng)價(jià)了抗氧化能力。測(cè)試了總黃酮和蘆丁的還原能力、抑制超氧陰離子自由基(O2-·)的能力、DPPH自由基的清除能力、羥基自由基的清除能力。結(jié)果表明:小黑藥總黃酮有良好的抗豬油氧化的作用,其抗光氧化的效果高于自動(dòng)氧化;還原能力、抑制超氧陰離子自由基(O2-·)的能力、DPPH自由基的清除能力略低于蘆丁,羥基自由基的清除能力高于蘆丁;小黑藥全株總黃酮含量為2.52%。

Antioxidant Activity of Total Flavonoids from Sanicula astrantiifolia Wolff

Abstract In this paper,the content and the optimum extraction of total flavonoid from Sanicula astrantiifolia Wolff were studied.The antioxidant activity of total flavonoids and the reference substance BHT were detected by Schaal Oven Method and UV photo-induced,and the antioxidant activity was estimated by inhibition ratio of POV.The reduction capacity of total flavonoids and Rutin,the inhibition ratio of total flavonoids to superoxide free radical(O2-·),its DPPH and hydroxyl free radicals-cavenging effect were determined.The results showed that:The total flavonoids from Sanicula astrantiifolia Wolff showed significant inhibitory effects on the oxidation of lard oil,its antioxidant effect was greater than automatic oxidation.The reduction capacity,the inhibition ratio to superoxide free radical(O2-·),its DPPH free radicals-cavenging effect were slightly lower than Rutin,and its hydroxyl free radicals-cavenging effect was greater than Rutin.The content of total flavonoid from Sanicula astrantiifolia Wolff was 2.52%.

小黑藥(Sanicula astrantiifolia Wolff)具有補(bǔ)肺益腎、治療肺結(jié)核、腎虛腰痛、頭昏等功效[1]。云南民間常食用小黑藥,以提高免疫力和鎮(zhèn)痛、止咳等[2]。現(xiàn)代食品工業(yè)中也通過(guò)小黑藥改善產(chǎn)品質(zhì)量,提高營(yíng)養(yǎng)價(jià)值,在油脂及相關(guān)產(chǎn)業(yè)中得到積極應(yīng)用[3-5]。筆者研究了小黑藥中的總黃酮含量、抗豬油氧化效果及抗氧化性能,旨在探索小黑藥中的活性成分及功能,為深入開(kāi)發(fā)和利用小黑藥提供依據(jù)。
1 材料與方法
1.1 試驗(yàn)材料
1.1.1 試材。小黑藥(曲靖農(nóng)貿(mào)市場(chǎng)購(gòu)買(mǎi));豬油(曲靖農(nóng)貿(mào)市場(chǎng)購(gòu)買(mǎi)新鮮豬肥膘,實(shí)驗(yàn)室火煉法提取);蘆丁(中國(guó)藥品生物制品檢定所);Tris(北京西美杰科技有限公司);1,1-二苯-2-苦基肼(日本和光純樂(lè)工業(yè)株式會(huì)社);二丁基羥基甲苯(BHT)(鄭州超凡有限公司);氯化亞鐵(天津市風(fēng)船化學(xué)試劑科技有限公司);……
