以衛(wèi)矛(Euonymusalatus(Thunb.) Sieb.)莖段為外植體,通過在4—9月不同時(shí)間取材,并采用升汞和次氯酸鈉不同處理時(shí)間、升汞結(jié)合吐溫、紫外線照射等消毒方法,篩選外植體最佳取材時(shí)間和最佳消毒方法;同時(shí),研究了不同質(zhì)量濃度的激素和瓊脂對(duì)不定芽誘導(dǎo)的影響,并采用L9(34)正交試驗(yàn)設(shè)計(jì),探討MS、B5、White不同培養(yǎng)基和不同質(zhì)量濃度的6-BA、NAA、GA3對(duì)不定芽增殖的影響,篩選出不定芽誘導(dǎo)和增殖的最佳培養(yǎng)基。結(jié)果表明:5月中旬為莖段的最佳取材時(shí)間;HgCl28 min并加入兩滴吐溫-20為消毒的最佳方法;MS+6-BA2.0 mg/L+NAA0.05 mg/L+瓊脂7.0 g/L是衛(wèi)矛莖段不定芽誘導(dǎo)的最佳培養(yǎng)基,誘導(dǎo)率高達(dá)70.67%;在正交試驗(yàn)的9個(gè)處理中,以MS+6-BA0.5 mg/L+NAA0.05 mg/L的增殖系數(shù)最高為19.25,通過極差分析得出MS+6-BA1.0 mg/L+NAA0.05 mg/L是衛(wèi)矛不定芽增殖的最佳培養(yǎng)基,增殖系數(shù)為23.51。

We used theEuonymusalatusstem segment as explants to study the effects of different sampling time on the disinfection of explants between 4 to 9 months, and the effects of different disinfection methods on the disinfection of stem segment, including mercuric chloride and sodium hypochlorite different processing time. We also studied the effect of hormone and agar in different concentrations on adventitious bud induction. Through L9(34) orthogonal test, we studied the effect of the MS, B5, White, and different concentration of 6-BA, NAA and GA3on the adventitious buds proliferation, and we finally chose out the optimum culture medium for adventitious bud induction and proliferation. Mid-May is the best sampling time ofEuonymusstem segments; mercuric chloride combined with two drops of tween-20 for 8 min is the best method forEuonymusstem segments sterilization; MS+6-BA 2.0 mg/L+NAA 0.05 mg/L+agar 7.0 g/L is the best culture medium forEuonymusstem segments of adventitious bud induction, adventitious bud induction rate reaches 70.67%; Among nine procedures in orthogonal test, the proliferation coefficient is the highest (19.25) in the first processing. MS+6-BA 1.0 mg/L+NAA0.05 mg/L is the optimum culture medium for adventitious bud proliferation inEuonymuswith the proliferation coefficient of 23.51.
衛(wèi)矛(Euonymusalatus(Thunb.) Sieb.)是衛(wèi)矛科(Celastraceae)衛(wèi)矛屬(EuonymusL.)落葉灌木,又名鬼箭羽、見腫消、千層皮等。其株形密集,秋葉紫紅色,假種皮橘紅色,極具觀賞價(jià)值,為重要的野生觀葉觀果的灌叢。同時(shí),衛(wèi)矛根、皮均可入藥,具有破血、殺蟲、通經(jīng)、散疲止痛等功效和調(diào)血脂、降血糖、延緩動(dòng)脈粥樣硬化等作用,近年來(lái)我國(guó)臨床上用于治療糖尿病已取得顯著效果[1],衛(wèi)矛在韓國(guó)和日本也多用于對(duì)抗腫瘤[2-3],具有很好的藥用價(jià)值。現(xiàn)如今衛(wèi)矛已在歐洲普遍栽培,中國(guó)雖然除新疆、青海、西藏、廣東及海南以外,各地均有分布,但是種植卻不普遍,還有待開發(fā)培育。……
