摘 要:為了獲得正常形態的冰燈玉露組培移栽苗,以冰燈玉露再生苗為外植體,采用含有不同激素的培養基、不同的培養溫度及光照強度等方法,以找到適宜的促成其組培苗正常形態建成的培養條件。結果顯示,采用MS+0.25 mg·L-1 BA培養基,在20 ℃的培養溫度、3 000 lx的光照強度、24 h連續光照條件下,可以獲得生長形態正常(與品種特性相一致)的組培苗。在此條件下,可以減少組培快繁中冰燈玉露畸形苗的產生,同時也會縮短冰燈玉露組培苗移栽成苗的時間。

Study on the Influence Factors of Morphogenesis of Haworthia cooperivar Tissue Culture Seedling

Abstract: In order to obtain the normal form of Haworthia cooperivar seedlings, the regeneration plantlets of Haworthia cooperivar were used as explants, using the culture mediums containing different hormones, different culture temperature and light intensity, the suitable culture condition for normal morphogenesis of plantlets was found. The results showed that the growth form of tissue culture seedlings was same as the seedling by ramet propagation under the condition of at 20 ℃ culture temperature, using the light intensity of 3 000 lx, and continuous illumination 24 h, on the medium MS+0.25 mg·L-1 BA. Under these conditions, the production of malformation Haworthia cooperivar tissue culture seedling will be reduced and the time of plantlet formation of Haworthia cooperivar tissue culture seedling will be shorten.

近年來,采用組培快繁技術對多肉植物進行繁殖的研究越來越多,并已在生產中得到了廣泛的應用[1-9]。隨著這一繁殖技術的推廣與使用,生產者發現,通過組織培養繁殖的商品苗在外觀形態上與分株或播種繁殖苗存在明顯的差異[10-15],其中,組培苗基部葉片較長,畸形,觀賞效果差,認為對這些葉片去除后會導致植株整體形態不完整,從而失去觀賞價值。其原因主要是由于移栽出的組培苗形態與自然繁殖苗不同,組培苗一般葉片細長,厚度不足,整體植株細弱,生長勢不強,因此導致組培苗需要較長時間的生長,新生葉才能恢復其正常的品種特征,不僅影響了這些多肉植物的商品價值,也增加了培養時間、提高了生產成本,從而影響了這一技術的應用前景。
