野生桑葚中花色苷成分分析

運用固相萃取純化技術與高效液相色譜/二極管陣列檢測器/電噴霧質譜聯用技術,采用Zorbax SB-C18色譜柱(250mm×4.6mm,5μm),甲醇-5%甲酸水溶液為流動相,流速為1.0mL/min,檢測波長為520nm,以矢車菊素3-葡萄糖苷為對照,外標法測定了野生桑葚花色苷含量,并通過紫外掃描光譜和電噴霧質譜正離子碎片信息確定了花色苷的成分組成。結果表明:野生桑葚總花色苷含量為154.27mg/100g,含有的三種花色苷成分分別為矢車菊素3-葡萄糖苷、矢車菊素3-蕓香糖苷和天竺葵素3-葡萄糖苷,其相對含量為67.52%、31.29%和1.06%。

Abstract:To establish a method to determine anthocyanin content and composition in wild mulberry,solid phase extraction(SPE)and high performance liquid chromatography equipped with a DAD detector and electro-spray ionization mass spectrometry(HPLC/DAD/ESI/MS)were used.The analysis was carried on Zorbax SB-C18column(250mm ×4.6mm,5μm).The mobile phase was aqueous 5%formic acid solution and methanol at the flow-rate of 1.0mL/min,the detection wavelength was 520nm.The content of gross anthocyanins was 154.27mg/100g by HPLC expressed as milligrams of cyanidin 3-glucoside equivalent per 100g of fresh weight.Three anthocyanins in wild mulberry were identified by UV-Vis and ESI+-MS spectra which were cyanidin 3-glucoside,cyanidin 3-rutinoside and pelargonidin 3-glucoside,the relative content of which were 67.52%,31.29%,1.06%respectively.

桑葚(Mulberry)是桑科桑屬落葉喬木桑樹Morusalba Linn.的果實,曬干或蒸后曬干可入藥,味甘酸,性寒,歸心、肝、腎經,中醫可用于治療肝腎不足和血虛精虧導致的頭暈目眩、腰酸耳鳴、失眠多夢、津傷口渴等癥。桑葚酸甜適口,營養豐富,富含鞣酸、蘋果酸、多種維生素和人體必需的氨基酸及鋅、鉀、鎂、磷等微量礦質元素,具有良好的保健功能,很早就被當作水果和中藥材加以應用[1]。此外,桑葚顏色紫紅,含有大量的天然紅色素。桑葚紅色素屬花色苷類成分,是果蔬及其產品呈現從紅色到藍色的物質基礎,具有抑制血小板凝固,預防血栓、心臟病,抗癌,延緩衰老等作用[2-4]。……
