毛果楊PtAREB1基因啟動子的克隆及功能1)

摘要為了研究楊樹ABF2同源基因的表達規律,從毛果楊基因組DNA中克隆出PtAREB1基因上游一段1 800 bp序列。序列分析結果表明,該序列含有逆境脅迫響應元件TC-rich repeats、ABA應答元件ABRE和茉莉酸甲酯(Methyl Jasmonate,MeJA)應答元件TGACG-motif等脅迫相關元件。在序列分析的基礎上,構建了PtAREB1基因啟動子驅動GUS報告基因的植物表達載體,利用農桿菌介導的花粉管通道法獲得轉基因擬南芥。結果表明PtAREB1啟動子可以在干旱、ABA、鹽、MeJA和SA脅迫下,驅動GUS基因在轉基因擬南芥的根、莖和葉中表達。說明PtAREB1基因可能與干旱、高鹽等脅迫應答緊密相關。

In order to study the expression and regulation of ABF homologous gene in poplar, a 1 800 bp 5’flanking sequence ofPtAREB1 gene was isolated by PCR from genomic DNA ofPopulustrichocarpa. By promoter sequence analysis, the sequence contained stress-response element TC-rich, ABA-response element ABRE and MeJA-response element TGACG-motif. By sequence analysis, thePtAREB1 promoter was fused to the GUS reporter gene to characterize its expression pattern inP.trichocarpa. From theAgrobacteriummediated transformation ofArabidopsisthaliana, the GUS gene was induced inArabidopsisthaliana, and it expressed in roots, stems and leaves under ABA, drought, high salt, MeJA and SA, suggesting thePtAREB1 promoter was responsive to ABA, drought and high salt stress.

脫落酸(abscisic acid,ABA)是一種非常重要的植物激素,它的主要作用是在脅迫下對植物營養組織適應性有基礎調節作用,如干旱和高鹽等[5-6]。在逆境下轉錄因子作用于啟動子調控下游基因上調或下調的表達[7],大多數受ABA誘導的基因在啟動子區域都有保守序列(C/T)ACGTGGC序列ABRE(ABA responsive element)順式作用元件[8]。ABA反應元件結合蛋白/AREB結合因子(AREB/ABF)基因亞家族包含在ABA介導的信號通路中的關鍵轉錄因子。ABFs(ABRE blinding factors)可以在非生物脅迫下激活ABRE相關基因的表達,提高植物非生物脅迫耐受性。
近幾年來,在模式植物研究中ABF轉錄因子已成為熱點。ABF基因在諸多非生物脅迫中被特異性誘導表達。……
