響應面法優(yōu)化Ganoderma lucidum產(chǎn)漆酶種子培養(yǎng)基

以菌絲體生物量為指標,采用響應面法對產(chǎn)漆酶的靈芝菌UIM281的種子培養(yǎng)基組成進行了研究。首先采用單因素實驗確定影響UIM281菌絲體生長的碳源、氮源及無機鹽的種類和濃度范圍。在此基礎(chǔ)上,應用Box-Behnken設(shè)計對影響菌絲體生物量的顯著因素進行優(yōu)化,最終建立了以菌絲體生物量為響應值的二次回歸方程模型,獲得了最適的菌絲體增殖種子培養(yǎng)基。結(jié)果表明,在VB10.1g/L的條件下最佳種子培養(yǎng)基組成為:玉米粉18.0g/L,豆餅粉12.3g/L,MgSO4·7H2O 1.8g/L,在28℃,150r/min條件下振蕩培養(yǎng)6d后,可獲得24.18g/L菌絲體,可為漆酶發(fā)酵培養(yǎng)提供質(zhì)優(yōu)量大的接種體。

Abstract:Targeting on mycelial biomass,response surface methodology(RSM) was applied to study the seeding medium components of UIM281,a Ganoderma lucidum strain producing laccase.Firstly,the categories and concentration scales of carbon source,nitrogen sources and inorganic salt affecting the growth of UIM281 mycelium were confirmed by single factor experiments.Based on this result,Box-Behnken design was used to optimize significant factors affecting mycelial biomass.Finally,the second-order equation model for mycelial biomass of UIM281 was established and seeding medium suitable to mycelial proliferation was obtained.The results showed when VB1was prior designated 0.1g/L,the optimal composing for seeding medium was as following:18.0g/L of corn flour,12.3g/L of soybean powder,1.8g/L of MgSO4·7H2O.When cultivated at 28℃and 150r/min for 6d,24.18g/L of biomass was obtained,which provided the excellent inoculum for the laccase fermentation.

漆酶是一種含銅的多酚氧化酶,在食品加工、造紙、廢水處理等方面具有廣泛的應用價值。靈芝菌(Ganoderma lucidum)除富含多糖、三萜類和肽類等有效生物活性成分外,也是產(chǎn)漆酶的重要真菌。在真菌漆酶發(fā)酵領(lǐng)域,高產(chǎn)菌株的選育、發(fā)酵工藝及產(chǎn)酶條件的優(yōu)化已開展了很多研究[1-2]。漆酶發(fā)酵培養(yǎng)主要采用兩種接種方式:一是采用從斜面或平板上挑取菌片直接接入發(fā)酵培養(yǎng)基進行漆酶發(fā)酵[3-4],二是采用經(jīng)種子擴培后再接入發(fā)酵培養(yǎng)基中進行產(chǎn)酶[5-6];……
