


[摘要] 目的 探討急性冠脈綜合征患者外周血微小RNA-208(miR-208)的表達及其與炎性因子表達水平的關系。方法 整群選取2014年4月—2015年5月大理大學附屬醫院心血管內科收治的138例ACS患者血清臨床資料,以 52名健康體檢者為對照組。分析miR-208在ACS患者血清中的表達及其與炎性因子TNF-α、CRP、IL -6的關系。結果 與對照組相比,miR-208在ACS患者血清中的表達升高2-ΔΔCt:(24.28±3.85) vs (7.83±1.54) P<0.001,并且miRNA-208表達的升高和炎癥因子分泌呈明顯正相關TNF-α (r=0.612)、CRP (r=0.443),IL -6 (r=0.543) P<0.05。 結論MiR-208表達在AMI患者血清中上調,并與炎癥相關,其可能成為ACS新的潛在診療靶點。

[Abstract] Objective To investigate the expression of RNA-208 (miR-208) in peripheral blood of patients with acute coronary syndrome and its relationship with the expression of inflammatory cytokines. Methods Group selection the clinical data of patients with ACS admitted to Department of Cardiovascular Medicine,The Affiliated Hospital of Dali University from April 2014 to May 2015 were enrolled.138 ACS patients and 52 healthy persons with matched age and gender served as healthy control group. The levels of serum miRNA-208, IL-6, CRP, TNF-αin all the subjects were recorded. The correlation between serum miR-208 and the inflammatory cytokines was analyzed. Results Compared with control group, miR-208 expression in ACS patients was higher (2-ΔΔCt (24.28±3.85) vs (7.83±1.54), P<0.001. MiR-208 expression was associated with inflammation factorsTNF-α (r=0.612),CRP (r=0.443),IL -6 (r=0.543), P<0.05. Conclusion Upregulated serum miR-208 is involved in the pathophysiology of ACS. MiR-208 may be used as a reference to predict, diagnosis and treat ACS.

動脈粥樣硬化(atherosclerosis,As)引起的心腦血管疾病發病率漸呈上升趨勢,已成為危害人類健康的主要殺手[1]。對其發病機制,多數學者支持炎癥反應學說,認為血管壁的慢性炎癥是導致As發生發展的重要因素。在動脈粥樣硬化性疾病中,急性冠脈綜合征(acute coronary syndrome, ACS)是一組由急性心肌缺血引起的綜合征,炎癥因子激活在其中發揮重要的作用[2]。MiRNAs是一類內源性、非編碼小RNA序列。MiRNA基因先在細胞核內轉錄出初始miRNA,再經Drosha- DGCR8復合物剪切為60~80堿基長度帶有莖-環結構的miRNA[3]。它們主要通過與蛋白編碼基因外顯子區或基因信使RNA(mRNA)的種子序列互補結合,致靶基因降解和/或翻譯抑制[4]。近年來,隨著分子生物學及高通量技術的發展,miRNA 在As中的作用逐漸引起人們重視。該研究通過檢測2014 年4 月—2015年5月大理大學附屬醫院心血管內科收治的138 例ACS患者血清miR-208表達水平,探討其與炎癥因子相互關系,以期為ACS診療提供新的靶點,報道如下。……
