【摘要】目的:探討HMGA2在睪丸癌組織中的表達(dá)及意義。方法:運(yùn)用熒光定量PCR(Q-PCR)和Western blot 方法檢測HMGA2mRNA和蛋白在17例睪丸腫瘤和15例正常睪丸組織中的表達(dá)差異、免疫組織化學(xué)(IHC)方法檢測HMGA2蛋白在44例睪丸腫瘤,30例良性組織芯片中的表達(dá)差異。結(jié)果:HMGA2mRNA表達(dá)在癌組織中明顯高于良性組織:(9.09±2.95) vs. (3.86±1.66), P=0.036、HMGA2蛋白在癌腫組織中的表達(dá)亦顯著高于良性組織:(9.27±2.91) vs. (4.66±1.62), P=0.037。 HMGA2蛋白的表達(dá)高低和睪丸腫瘤的臨床分期及腫瘤大小密切相關(guān),P<0.05。結(jié)論:HMGA2在睪丸腫瘤組織中上調(diào)表達(dá)并和腫瘤進(jìn)展相關(guān),有望成為睪丸腫瘤治療的分子靶點(diǎn)。

【Abstract】Objectives: To investigate the expression and significance of HMGA2 in testicular cancer tissue. Methods: Using the fluorescent quantitative PCR (Q - PCR) and Western blot methods to detect HMGA2mRNA and protein in 17 cases of testicular tumors and 15 cases of normal testicular tissue, immunohistochemical (IHC) method to detect HMGA2 protein in 44 cases of testicular tumors and 30 cases of benign tissue by microarray. Results: HMGA2mRNA expression in cancerous tissue was significantly higher than that of benign group (9.09 ±2.95 VS 3.86± 1.66, P = 0.036); HMGA2 protein expression in tumor tissue was significantly higher than that of benign organization (9.27±2.91 VS 4.66 ± 1.62, P = 0.037). HMGA2 protein expression levels in testicular tumors were associated with clinical staging and the tumor size (P<0.05). Conclusion: Up-expressed of HMGA2 in testicular cancer is associated with tumor progression and size, which is expected to become the molecular targets of testicular tumor treatment.

睪丸腫瘤是青年男性最常見的惡性實(shí)體腫瘤,隨著腫瘤綜合治療的進(jìn)展,睪丸癌的治愈率也得到了很大程度的提高。但對于臨床進(jìn)展期及轉(zhuǎn)移性腫瘤,臨床治療效果仍舊較差,尋找合適的腫瘤進(jìn)展期腫瘤標(biāo)記物以及如何評估患者的預(yù)后風(fēng)險(xiǎn)是臨床上亟待解決的問題。
高遷移率蛋白A2(HMGA2)是在腫瘤中廣泛表達(dá)的蛋白之一,其表達(dá)異常與人類多腫瘤的發(fā)生、發(fā)展及預(yù)后相關(guān)。目前HMGA2與睪丸腫瘤組織的相關(guān)性國內(nèi)較少研究,我們在這方面做了一定研究,報(bào)道如下。
1材料和方法
1.1材料
收集近5年來醫(yī)院就診的17例睪丸腫瘤活檢或者手術(shù)組織標(biāo)本以及15例正常睪丸活檢組織標(biāo)本用于Q-PCR及WB實(shí)驗(yàn),其中腫瘤患者均未接受治療,初次確診為睪丸腫瘤,病理類型不限,所有標(biāo)本均經(jīng)過最后的病理確認(rèn)。組織來源取材時(shí)候均向患者說明,并簽署知情同意書。免疫組化(IHC)組織芯片標(biāo)本包括44例睪丸腫瘤標(biāo)本(病理及臨床分期不限)及30例良性對照標(biāo)本。……
