[摘要] 目的 研究高遷移率族蛋白1(HMGB1)在胃癌組織的表達(dá),以及其與相關(guān)胃癌臨床病理特征的關(guān)系。方法 選取內(nèi)蒙古醫(yī)科大學(xué)第一附屬醫(yī)院2012年3月~2013年9月術(shù)后病理明確診斷的104例胃癌及其癌旁組織標(biāo)本,應(yīng)用免疫組織化學(xué)法檢測(cè)標(biāo)本HMGB1蛋白的表達(dá),分析HMGB1在胃癌組織和癌旁組織的差異及其與胃癌臨床病理特征關(guān)系。檢測(cè)HMGB1 mRNA在胃癌細(xì)胞系SGC-7901、N87、MKN45及正常胃黏膜細(xì)胞系GES-1中的表達(dá)。 結(jié)果 免疫組化顯示:HMGB1陽(yáng)性顆粒不僅位于細(xì)胞核上,還位于細(xì)胞質(zhì)中;胃癌組織中的表達(dá)水平高于胃癌的癌旁組織(P < 0.01)。QPCR均顯示:HMGB1 mRNA在正常胃黏膜細(xì)胞中的表達(dá)水平低于低分化胃癌細(xì)胞(P < 0.01)。 結(jié)論 HMGB1在胃癌組織中表達(dá)較高,特別是低分化胃癌,推測(cè)可能參與胃癌發(fā)生和發(fā)展,但在胃癌浸潤(rùn)轉(zhuǎn)移中的臨床意義需要擴(kuò)大樣本量進(jìn)行進(jìn)一步研究。

Expression of HMGB1 in gastric cancer and its relationship with clinical pathological characteristics

[Abstract] Objective To study the expression of HMGB1 in gastric carcinoma and the relationship between the expression and clinicopathological features of gastric cancer. Methods From March 2012 to September 2013, in the First Affiliated Hospital of Inner Mongolia Medical University, 104 samples of postoperative pathological diagnosis gastric cancer and their adjacent tissues were selected. Immunohistochemical method was used to detected HMGB1 protein expression, differences of HMGB1 in gastric cancer tissues and adjacent tissues, and its relationship with clinical pathological features of gastric carcinoma were analyzed, the expression of mRNA HMGB1 in gastric cancer cell line SGC-7901, N87, MKN45 and normal gastric mucosal cell line GES-1 were detected. Results Immunohistochemical staining showed that HMGB1 positive particles not only were located in the nucleus, but also in the cytoplasm, and its expression level in gastric cancer tissue was higher than that in gastric cancer adjacent tissue (P < 0.01). QPCR showed that HMGB1 mRNA in normal gastric mucosal cells was lower than that in poorly differentiated gastric carcinoma cells (P < 0.01). Conclusion HMGB1 is highly expressed in gastric cancer tissues, especially in poorly differentiated gastric carcinoma and speculated that it may participate in the process of the occurrence and development of gastric cancer, but in gastric cancer invasion and metastasis and its clinical significance need to expand the sample size for further study.
