[摘要] 目的 探討心肌內基質金屬蛋白酶及組織基質金屬蛋白酶抑制因子表達變化。方法 2017年1—2月隨機選取該院實驗室20只SD大鼠,依據隨機數字表法將其分為阿霉素組和對照組兩組,每組10只,應用半定量RT-PCR法測定mRNA表達水平,運用Western blotting方法對蛋白質水平進行檢測,然后對心肌組織MMP-2、MMP-9表達、TIMP-1、MAPKs活性受到阿霉素的影響進行分析。結果 心肌組織MMP-2、MMP-9mRNA在阿霉素的作用下提升,心肌組織MMP-2、MMP-9蛋白表達水平在阿霉素處理的情況下提升;心肌組織TIMP-1mRNA水平會在阿霉素的作用下提升,心肌組織TIMP-1蛋白水平在阿霉素處理的情況下提升;心肌組織p38MAPK磷酸化水平在阿霉素處理的情況下提升,而ERK、JNK MAPK MAPKs磷酸化水平并沒有在阿霉素處理的情況下提升。結論 心肌內基質金屬蛋白酶及組織基質金屬蛋白酶抑制因子表達受到阿霉素的直接而深刻的影響,值得臨床充分重視。

Changes of Intramyocardial Matrx Metalloproteinases and Tissue Inhibitor of Metalloproteinase Protein Expression

[Abstract] Objective To study the changes of intramyocardial matrx metalloproteinases and tissue inhibitor of metalloproteinase protein expression. Methods 20 SD rats in the laboratory in our hospital from January to February 2017 were randomly selected and divided into the Adriamycin group and the control group, the mRNA expression level was measured by the semi-quantitative RT-PCR method, while the protein level was tested by the Western blotting method, and then the effect of adriamycin on the cardiac muscle tissue MMP-2, MMP-9 expression, TIMP-1 and MAPKs activity was analyzed. Results The cardiac muscle tissue MMP-2 and MMP-9mRNA increased affected by the adriamycin, and the cardiac muscle tissue MMP-2 and MMP-9 protein expression level increased under the treatment of adriamycin, and the cardiac muscle tissue TIMP-1mRNA level increased affected by the adriamycin, the cardiac muscle tissue TIMP-1 protein level increased under the treatment of adriamycin, and the cardiac muscle tissue p38MAPK phosphorylation level increased under the treatment of adriamycin, but the ERK and JNK MAPK MAPKs phosphorylation levels did not increase under the treatment of adriamycin,. Conclusion Adriamycin has a direct and deep effect on the intramyocardial matrx metalloproteinases and tissue inhibitor of metalloproteinase protein expression, which is worth full attention in clinic.

心力衰竭屬于一組綜合征,在臨床極為常見,心室重塑及心肌損傷是其發病及發展的主要機制,極易降低心功能[1]。阿霉素屬于一種廣譜高效的抗腫瘤藥物,在腫瘤的治療中應用極為廣泛,但是由于其毒性會損傷心肌組織,因此其在臨床的應用受到了一定程度的限制。充血性心力衰竭的心肌病是其主要的毒性作用,患者具有極差的預后[2]。在對完整的心肌結構進行保持的過程中,細胞外基質發揮著極為重要的作用。……
