吲達帕胺片含量測定方法改進研究

目的對吲達帕胺片的含量測定方法進行研究，找出分析方法的關鍵控制點。方法 按照2015年版《中國藥典（二部）》吲達帕胺片含量測定方法，將吲達帕胺片研細后的粉末全部過6號篩，加乙醇適量，水浴加熱溶解一定時間，按紫外－可見分光光度法[2015年版《中國藥典(四部)》（通則0401）]，在242 nm波長處測定吸光度。結果 吲達帕胺片的含量在90．0% ～110．0%之間，符合《中國藥典》的要求。結論 經過改進的測定方法能確保在不同實驗室、不同型號的儀器，以及不同分析人員等影響因素下，始終能準確地檢測出吲達帕胺片的含量。

Abstract：Ob jective To study the method for content determination of Indapamide Tablets，and to find out the critical point control of analytical methods．M ethods According to the method of China Pharmacopoeia（2015 Edition，VolumeⅡ）to determine the content of Indapamide Tablets，the porphyrization powder of Indapamide Tablets was over the No．6 sieve，the right amount of alcohol was added and heated up in water bath for a certain time．Absorbance was measured at the wavelength of 242 nm by ultraviolet－visible spectrophotometry of China Pharmacopoeia（2015 Edition，VolumeⅣ，General 0401）．Results The content of Indapamide Tablets ranged from 90．0% to 110．0%，which was in accordance with the requirements of Chinese Pharmacopoeia．Conclusion The improved method can always accurately determine the content of Indapamide Tablets in influence factor of different laboratories，different types of instruments and different analysts．

吲達帕胺為非噻嗪類吲哚衍生物，具有擴張血管平滑肌及利尿的作用[1－3]，是目前臨床廣泛應用的一線抗高血壓藥，可單獨用于輕、中、高度高血壓，降壓溫和，療效確切，為理想的長效降壓藥[4－10]。本公司曾安排不同的化驗人員按照2015年版《中國藥典(二部)》[11]含量測定項下方法檢測，結果含量差異較大。對生產的吲達帕胺片（批號為15110303）留樣檢測，不同的化驗人員檢測結果不同：新化驗員檢測含量為87．2%，不符合《中國藥典》標準90．0%～110．0%的要求，為不合格產品；有經驗的化驗員檢測含量為96．7%，符合《中國藥典》規定。最初認為是含量均勻度問題，后通過不同化驗員進行同批隨機抽檢，對含量均勻度進行檢測，結果發現，雖新老化驗員含量均勻度檢測值均在藥典規定范圍，但新化驗員10片中每1片含量均比老化驗員含量檢測低近10%，10片含量平均值也相差10%，從而排除含量均勻性問題。……
