大落新婦根中三萜類成分的研究

虎耳草科落新婦屬植物在全世界約有18 種,我國產7 種,大部分地區均有分布[1]。貴州黔東南一帶苗族地區以落新婦和大落新婦最常用,主要治療跌打損傷,骨疼痛、關節疼痛、風濕病、腰腿痛等,在抗炎、消腫、止痛、活血化瘀方面功效顯著。

現代研究表明,落新婦屬植物中主要化學成分為三萜、香豆素、黃酮,在鎮痛消炎、抗腫瘤、鎮咳平喘、抗菌、抗病毒、治療燒燙傷等方面有廣泛的生物活性[2-12]。其所含三萜是一種少見的羧基從C17位遷移到C14位的烏蘇烷型和齊墩果烷型,在抗腫瘤、抗真菌、平喘等方面有很好的作用[3,13]。

目前,尚未見對大落新婦化學成分的研究?;诖舐湫聥D的豐富資源、廣泛使用和顯著療效,以及具有潛在生物活性的特征三萜類,本實驗對其根進行研究,從中分離得到9 個化合物,均為首次從該植物中發現。

1 材料

Q-TOF 質譜儀(德國布魯克公司);Inova-400 MHz 核磁共振儀(美國瓦里安公司);R215 實驗室規模旋轉蒸發儀(瑞士步琪公司);FZ102 微型植物粉碎機(上海書培實驗設備有限公司);電子天平(瑞士Metter-Toledo 公司);Sephadex LH-20(美國通用電氣醫療集團);GF254制備薄層板及200~300 目柱色譜用硅膠(青島海洋化工有限公司)。所用試劑均為分析純。

藥材于2013 年8 月采自六盤水市鐘山區石龍村,經貴州中醫藥大學趙俊華教授鑒定為虎耳草科落新婦屬植物大落新婦Astilbe grandisStapf ex Wils.,標本存放于貴州中醫藥大學苗醫藥重點實驗室,標本號為20130814201。

2 提取與分離

將自然風干的14 kg 大落新婦根粉碎,加甲醇冷浸提取4 次,每次3 d,過濾提取液,減壓回收甲醇后得浸膏1.1 kg,蒸餾水溶解后經D101 大孔樹脂柱,分別用水、50% 乙醇、90% 乙醇梯度洗脫,得到90%乙醇部分(518 g),進行硅膠柱層析,二氯甲烷-甲醇(100∶1~8∶2) 梯度洗脫,得到3 個組分Fr.A~C。Fr.B 經硅膠柱層析,石油醚-乙酸乙酯(100∶1~1∶1) 梯度洗脫,得到5個組分Fr.B1~Fr.B5。Fr.B2 經硅膠柱層析,二氯甲烷-乙酸乙酯 (100∶1~8∶2) 反復柱層析,Sephadex LH-20 純化,得到化合物3 (26 mg)、4(6 mg)、7 (101 mg);Fr.B3 經硅膠柱層析,二氯甲烷-乙酸乙酯(8∶2) 系統洗脫,析出白色粉末,過濾得到化合物8 (900 mg),母液部分再經硅膠柱層析,石油醚-乙酸乙酯(8∶2) 反復柱層析,Sephadex LH-20 純化,得到化合物1 (6 mg)、2(11 mg);Fr.B4 經硅膠柱層析,二氯甲烷-甲醇(100∶1~9∶1) 反復柱層析,Sephadex LH-20 純化,得到化合物5 (20 mg)、6 (38 mg)、9 (20 mg)。

3 結構鑒定

4 討論

本研究首次從大落新婦中分離得到羧基從C17位遷移到C14位的齊墩果烷型三萜,初步探明其主要成分類型,豐富了落新婦屬植物相關化合物的數量。下一步,將繼續對所得三萜進行生物活性考察,以期探明其作用機制,為后續研究提供依據。
