[摘要]目的 采用發(fā)酵法從冬蟲夏草發(fā)酵液及菌絲體中提取、分離蟲草多糖。 方法 采用乙醇沉淀法分離冬蟲夏草發(fā)酵液和菌絲體中的多糖;采用苯酚硫酸法測定樣品中多糖含量。 結(jié)果 采用此法進行制備、檢測,方法簡單快速,結(jié)果準(zhǔn)確,適宜推廣使用。結(jié)論 此法無需多糖純品,具有快速簡單、精確度高、靈敏度高、重現(xiàn)性較好的優(yōu)點,是檢測蟲草多糖含量的較優(yōu)方法之一。

[Abstract] Objective Using the method of inoculated fermentation to extract and separate the Cordyceps polysaccharide from the zymotic fluid and mycelium of Chinese caterpillar fungus. Methods Separated the polysaccharide of Chinese caterpillar fungus' zymotic fluid and mycelium in ethanol precipitation;survey and evaluate the polysaccharide content of samples in phenol-sulfuric acid method. Results It is profitable for using widely because of the fast, easy method and exact result for producing and detecting. Conclusion This method is one of excellent ways for detecting the Cordyceps polysaccharide contents and it doesn't need the polysaccharide from pure product. The advantages are fast and easy, high accuracy and sensitivity,and good reproducibility.

冬蟲夏草首載于本草從新,味甘,性溫,歸肺、腎經(jīng),具補肺益腎,止血化痰之功效。冬蟲蟲草中含量最高的活性物質(zhì)是蟲草多糖,因其可以抗氧化、抗腫瘤和增強免疫力等功效而受廣大人群青睞,由于天然的活性成分來源有限,現(xiàn)多采用人工發(fā)酵法生產(chǎn)制備。作者用苯酚-硫酸法測定蟲草多糖含量。
1 材料
1.1 供試材料
蟲草菌株(青海?。徽麴s水(雙蒸水)、發(fā)酵液精多糖Ⅰ和菌絲體精多糖Ⅱ;葡萄糖標(biāo)準(zhǔn)溶液(100 μg/mL)等。
1.2 儀器及設(shè)備
培養(yǎng)箱(常州市恒久儀器制造有限公司光照培養(yǎng)箱);高壓蒸汽滅菌設(shè)備(雞西辰豐醫(yī)療器械制造有限公司YXQ.L-75型立式壓力蒸汽滅菌器);大容量離心機(北京時代北利離心機有限公司GTR10-2);紫外分光光度計(上海悅豐儀器儀表有限公司)等。
1.3 培養(yǎng)基配方
1.3.1 活化培養(yǎng)基(g/mL) 蔗糖溶液1.30%,葡萄糖溶液1.30%,蛋白胨0.01%,酵母粉0.065%,瓊脂0.5%,KH2PO4 0.025%,MgSO4·7H2O 0.012 5%,pH自然,培養(yǎng)溫度27℃。
1.3.2 發(fā)酵培養(yǎng)基(g/mL) 葡萄糖1.75%,蛋白胨0.02%,酵母粉0.062 5%, KH2PO4 0.025%,MgSO4·7H2O 0.012 5%, pH自然,溫度27℃[1]。
