[摘要] 目的 探討3種不同劑量丹參酮ⅡA磺酸鈉對(duì)力竭運(yùn)動(dòng)后大鼠心肌的作用。 方法 建立大鼠力竭運(yùn)動(dòng)模型,30只大鼠隨機(jī)分為5組,A組:不做任何訓(xùn)練,常規(guī)飼養(yǎng),自由飲食;B組:力竭運(yùn)動(dòng)組;C組:力竭運(yùn)動(dòng)后注射低劑量丹參酮;D組:力竭運(yùn)動(dòng)后注射中劑量丹參酮;E組:力竭運(yùn)動(dòng)后注射高劑量丹參酮。實(shí)驗(yàn)動(dòng)物在訓(xùn)練5周后,取心肌組織,用透射電鏡觀察大鼠心肌結(jié)構(gòu)、局部和細(xì)胞器的變化。 結(jié)果 力竭運(yùn)動(dòng)可造成大部分心肌細(xì)胞的肌絲溶解,大量線粒體腫脹,一些區(qū)域的肌膜不完整,致使心肌細(xì)胞線粒體等細(xì)胞器外溢。而腹腔注射丹參酮低、中、高劑量組3組心肌纖維結(jié)構(gòu),細(xì)胞器較正常飼養(yǎng)組無明顯變化。 結(jié)論 丹參酮ⅡA磺酸鈉對(duì)力竭運(yùn)動(dòng)大鼠心肌有保護(hù)作用,其中以中劑量作用更明顯。

[Abstract] Objective To explore the different dose tanshinoneⅡA sodium on heart muscular tissues in rats after exhaustive exercise. Methods Rats exhaustion motion model was established. 30 rats were randomly divided into 5 groups including group A, group B, group C, group D, and group E. Group A didn't do any training and had the conventional breeding and free diet. Group B did the exhaustive exercise. Group C was injected low dose tanshinone after exhaustive exercise. Group D was injected middle dose tanshinone after exhaustive exercise. Group E was injected high dose tanshinone after exhaustive exercise. Experimental animals in training 5 weeks later, took the myocardial tissue, observed the changes of cardial structure and local and organelles of five groups by transmission electron microscope. Results Exhaustive exercise could lead to a majority of myocardial cell's myofibrillar melt, a large of mitochondria swell, some epimysiam was not integrate. Caused cell organ such as myocardial cell's myofibrillar overflew. But compared with group A, cell organ of intraperitoneal injection of low middle and high three different dose tanshinone had no obvious change. Conclusion Different dose tanshinone can protect myocardial injury after exhaustive exercise and the middle dose is the most effective.

研究已證實(shí)力竭運(yùn)動(dòng)可造成心肌損傷[1-2],對(duì)極少數(shù)運(yùn)動(dòng)員可造成運(yùn)動(dòng)猝死。本研究從新角度研究力竭運(yùn)動(dòng)造成心肌損傷的機(jī)理,尋求理想劑量丹參酮干預(yù)保護(hù)心肌細(xì)胞。在前期研究基礎(chǔ)上,進(jìn)一步研究如何更好地增強(qiáng)運(yùn)動(dòng)員的運(yùn)動(dòng)能力,以及減少力竭運(yùn)動(dòng)對(duì)心肌的損害的有效方法,采用不同劑量丹參酮分別干預(yù)力竭運(yùn)動(dòng)后的大鼠,旨在尋求干預(yù)保護(hù)心肌細(xì)胞的丹參酮最佳劑量,探討不同劑量丹參酮干擾力竭運(yùn)動(dòng)后大鼠的心肌恢復(fù)和保護(hù)作用。
1 材料與方法
