[摘要] 目的 探討趨化因子CCL4在非酒精脂肪性肝病(NAFLD)病人肝細胞中的表達水平,評價CCL4的表達水平是否與NAFLD的病情進展是否相關。方法 方便收集該院2013年1月—2015年11月間肝臟手術后分離培養的原代肝細胞39例,包括正常肝原代細胞13例、非酒精性脂肪肝病人肝細胞14例以及肝纖維化病人肝細胞12例,分別提取其細胞中總RNA樣品并進行逆轉錄后,通過Real-time PCR方法分析CCL4 mRNA的表達水平。結果 CCL4的表達水平在非酒精性脂肪肝(Steatosis)病人肝細胞中的表達水平為對照組的3.64倍;而在肝纖維化(Fibrosis)病人肝細胞中的表達水平則是對照組的33.69倍,是非酒精性脂肪肝組的9.25倍,差異均具有統計學意義(P<0.05)。 結論 CCL4的表達水平與非酒精性脂肪性肝病的發生及進展呈正相關,可考慮用作NAFLD診斷及治療的新靶點來進行進一步的開發研究。

The Expression Level of Chemokine CCL4 in the Primary Hepatocytes from NAFLD Patients

[Abstract] Objective To study the expression level of CCL4 mRNA in the primary hepatocytes from NAFLD patients, aiming to investigate the potential correlation between the expression level of CCL4 and the development of NAFLD. Methods We Convenient selection from January 2013 to November 2015 after liver surgery from 39 cases of primary cultured hepatocytes, including normal liver cells, 13 cases of patients with nonalcoholic fatty liver cells and liver cells in 14 cases of patients with liver fibrosis in 12 cases, the total RNA were extracted from the cells in the sample and for reverse transcription, expression analysis the level of CCL4 mRNA by Real-time PCR method. Results The expression level of CCL4 in steatosis group was 3.64 fold of that in normal group.Further, the expression level of CCL4 in fibrosis group were 33.69 fold and 9.25 fold of those in normal group and steatosis group, respectively. The differences between groups were all statistically significant(P<0.05). Conclusion The expression level of CCL4 was positively correlated with the development of NAFLD, making CCL4 a potential target for NAFLD prognosis and therapy.

非酒精性脂肪肝病 (Non-Alcoholic Fatty Liver Disease, NAFLD)與肥胖密切相關。近年來,NAFLD的發病率呈明顯的上升趨勢,目前已成為西方發達國家的第一大慢性肝病[1]。已經成為我國最常見的慢性肝病之一,其在人群中的發病率約為20%[2]。NAFLD可由單純的甘油三酯蓄積所表現出來的單純性脂肪肝(Steatosis),進一步發展成為脂肪性肝炎(Non-alcoholic Steotohepatitis, NASH),并可能最終發展為肝纖維化,肝硬化等疾病。在NAFLD病情進展中,肝臟中的炎癥反應起了重要的作用。Chemokine (C-C motif) ligand 4 (CCL4)是一種趨化因子,它具有募集自由殺傷細胞及單核細胞等免疫細胞的作用[3]。它在NAFLD病情進展中的作用尚未見報道。該研究方……
