[摘要] 目的 分析高同型半胱氨酸血癥對大鼠腦內NMDA受體亞單位蛋白表達的影響。方法 研究時間2015年7月—2016年4月,將36只SD大鼠隨機分為模型組(皮下注射用生理鹽水配成的蛋氨酸溶液)和對照組(相應體積的生理鹽水),檢測不同時間點大鼠血漿同型半胱氨酸含量及腦內NMDA受體亞單位(NR1、NR2A、NR2B)的蛋白含量。結果 模型組中第3小組大鼠的Hcy、腦內NMDA受體NR1、NR2A、NR2B分別為(21.46±1.37)、(0.99±0.13)、(0.95±0.18)、(1.47±0.25),均高于第2小組和第1小組(P<0.05),第1小組大鼠上述指標含量均為最低,分別為(10.14±1.13)、(0.74±0.11)、(0.70±0.23)、(0.68±0.13)。模型組3個小組大鼠的腦內NMDA受體NR1、NR2A、NR2B含量均高于對照組中對應小組大鼠,差異有統計學意義(P<0.05)。 結論 高同型半胱氨酸血癥會導致大鼠腦內NMDA受體亞單位的蛋白表達上升,加重大鼠腦神經損傷。

Effects of Hyperhomocysteinemia on NMDA Receptor Subunit Protein Expression in Rat Brain

[Abstract] Objective To investigate the effect of hyperhomocysteinemia on the expression of NMDA receptor subunit in rat brain. Methods Study time From July 2015 to April 2016,36 SD rats were randomly divided into a model group (the normal saline solution for subcutaneous injection)and a control group(the corresponding volume of saline),and the rats were sacrificed at different time points The plasma levels of homocysteine and the protein content of NMDA receptor subunits(NR1, NR2A, NR2B)in rats were determined. Results The levels of NMDA receptors NR1,NR2A and NR2B in the model group were (21.46±1.37),(0.99±0.13),(0.95±0.18),(1.47±0.25),(10.14±1.13),(0.74±0.11),(0.70±0.23),(0.68±0.13),respectively(P<0.05), and there was no significant difference between the group 1 and group 1.The contents of NMDA receptor NR1,NR2A,NR2B in three groups of rats in the model group were higher than those in the control group(P<0.05) the difference was scatistically significant. Conclusion Hyperhomocysteinemia can increase the protein expression of NMDA receptor subunits in the brain of rats and increase the damage of the cranial nerves.

高同型半胱氨酸血癥發病實質是由機體甲硫氨酸代謝障礙所引起的血漿或血清中游離及與蛋白結合的同型半胱氨酸、混硫化合物含量增高,目前已被證實是誘發心腦血管疾病的重要危險因素[1-2]。腦卒中是我國臨床常見的一種急性腦血管疾病,發生后不僅嚴重危害患者機體健康,同時也會給患者家庭造成沉重的經濟負擔[3]。高同型半胱氨酸血癥作為腦卒中獨立危險因素,對其誘發腦損害機制的深入研究,可為臨床制定能夠進一步改善腦卒中患者預后的治療方案提供參考依據[4]。基于上述現狀,該院于2015年7月—2016年4月選取36只實驗大鼠開展研究,針對高同型半胱氨酸血癥對大鼠腦內NMDA受體亞單位蛋白表達的影響進行了分析,現報道如下。……
