摘 要: 枯萎病是甜瓜等瓜類作物的毀滅性病害,F(xiàn)om-2基因介導(dǎo)了甜瓜抗枯萎病生理小種0和1,抗枯萎病育種是甜瓜等瓜類作物育種的重要目標(biāo)之一。通過同源克隆方法從薄皮甜瓜白啄瓜中克隆到了枯萎病抗性基因Fom-2,推定編碼1 099個氨基酸,所推定的蛋白質(zhì)序列經(jīng)Blast分析與數(shù)據(jù)庫中NBS-LRR類R基因具有較高的同源性。進(jìn)一步序列分析FOM-2蛋白具有典型的R基因的NB-ARC及SCOP 2個結(jié)構(gòu)功能域。實(shí)時(shí)熒光定量PCR結(jié)果表明,白啄瓜Fom-2基因經(jīng)枯萎病接種后表達(dá)明顯上調(diào)。白啄瓜Fom-2基因?yàn)榘鬃墓峡共∮N奠定了基礎(chǔ)。

Cloning and transcriptional analysis of resistance gene Fom-2 from Cucumis melo ssp. Agrestis Jeffrey

Abstract: Fusarium wilt (Fusarium oxysporum f. sp. melonis Snyder and Hansen) is one of destructive disease in melon crop production. Fom-2 gene was genetically identified to control the resistance of races 0 and 1. Resistance to fusarium wilt breeding is one of important targets for melon breeding. In this study, we cloned the Fom-2 gene from Baizhuogua melon (Cucumis melo ssp. agrestis Jeffrey) by using homological cloning method, which was deduced to encode 1099 amino acids and exhibited high similarity to NBS-LRR type of R genes in plant. The deduced FOM-2 protein contained NB-ARC and SCOP typical motifs of R gene. The expression of Fom-2 gene was highly induced after inoculation with fom race 1. In conclusion, the cloning of Fom-2 gene is good for breeding with resistance to fusarium wilt .

白啄瓜(Cucumis melo ssp. agrestis Jeffrey)屬于薄皮甜瓜類型,是溫州地區(qū)的特產(chǎn)瓜類作物,在溫州地區(qū)具有很大的栽培面積,栽培面積常年穩(wěn)定在600~700 hm2。同時(shí),溫州地區(qū)還具有接近我國人口最密集的長江三角洲蔬菜瓜果消費(fèi)市場的區(qū)位優(yōu)勢,當(dāng)天采摘,當(dāng)天即可送到市場,運(yùn)輸費(fèi)用低,白啄瓜等薄皮甜瓜生產(chǎn)具有非常強(qiáng)的市場競爭能力。近年來,隨著集約化、規(guī)模化的栽培,白啄瓜病蟲害擴(kuò)散蔓延十分迅速,對甜瓜產(chǎn)量和品質(zhì)造成極大的影響,尤其是枯萎病的影響。侵染甜瓜的枯萎病病原菌屬于甜瓜專化型尖孢鐮刀菌(Fusarium oxysporum f. sp. melonis Snyder and Hansen)。Risser等[1]認(rèn)為,可將尖孢鐮刀菌甜瓜專化型的病原菌劃分為4個生理小種,分別為race 0、 race 1、race 2和race 1, 2;3個抗病基因,分別為Fom-1、Fom-2和Fom-3,其中:Fom-1抗race 0和2,感race 1和race 1, 2;Fom-2抗race 0和1,感race 2和race 1, 2;Fom-3抗race 0, 1和2,感race 1, 2,且都是顯性基因[2]。Tarek等[3]根據(jù)文庫及Wang等[4]得到與Fom-2緊密連鎖的分子標(biāo)記,通過圖位克隆方法克隆了基因Fom-2,這是到目前為止,甜瓜中唯一克隆到的抗枯萎病基因。……
