不同濃度依托咪酯誘導(dǎo)人肝癌HepG2細(xì)胞體外凋亡的研究

[摘要] 目的 探討不同濃度的依托咪酯是否具有直接誘導(dǎo)人肝癌HepG2細(xì)胞凋亡的作用,尋找依托咪酯臨床治療的新途徑。 方法 人肝癌細(xì)胞株HepG2體外擴(kuò)增培養(yǎng)備用,選取不同濃度的依托咪酯E1、E2、E3和對(duì)照組(空白),分別培養(yǎng)12h、24h、48h后觀察。 結(jié)果 與空白對(duì)照組相比,培養(yǎng)12h、24h、48h后E1組、E2組人肝癌細(xì)胞株HepG2細(xì)胞凋亡率無(wú)明顯差異E3組凋亡率不斷增加,E1與E3組對(duì)比人肝癌細(xì)胞株HepG2細(xì)胞凋亡率具有顯著性差異E2與E3組對(duì)比人肝癌細(xì)胞株HepG2細(xì)胞凋亡率具有明顯差異結(jié)論 依托咪酯在臨床安全有效的濃度下不直接誘導(dǎo)人肝癌HepG2細(xì)胞的體外凋亡,濃度及時(shí)間達(dá)到一定峰值時(shí)可具有直接誘導(dǎo)人肝癌HepG2細(xì)胞體外凋亡的作用。

[Abstract] Objective To explore the different concentrations of etomidat whether directly induce the cell apoptosis to seek open up new ways of etomidate clinical treatment. Methods Human liver HepG2 cell line were amplificated cultivation for use in vitro, choose different concentrations of etomidate E1, E2 and E3 (blank) as control group.HepG2 were observed respectively after 12h, 24h, and 48h.With liver cancer HepG2 cell lines as target cells. Results Compared with the blank control group, 12h, 24h, 48h after culture E1 and E2 group liver HepG2 cell line apoptosis rate has no obvious difference(P>0.05), E3 group of apoptosis rate increased, and with significant difference (P<0.05); As the extension of incubation time and concentration increasing, E1 and E2 group compared with liver HepG2 cell line apoptosis rate have no significant difference(t=1.732,P>0.05), E1 and E3 group compared with liver HepG2 cell line apoptosis rate have significant difference(t=1.864,P<0.05),E2 and E3 group compared with liver HepG2 cell line apoptosis rate have obvious difference (t=1.691, P<0.05). Conclusion Etomidate with safe and effective concentrations in clinical are not directly induced HepG2 liver cancer cell apoptosis in vitro, but when concentration and time reach a certain peak can be directly induced the hepatocellular carcinoma HepG2 cells apoptosis in vitro.

依托咪酯可以直接抑制HL-60的細(xì)胞活性,并且誘發(fā)該細(xì)胞的凋亡,也有相關(guān)文獻(xiàn)報(bào)道依托咪酯對(duì)鼠肝線粒體能量代謝有影響,還有對(duì)肝臟缺血-再灌注損傷的保護(hù)均有作用[1-4],那么依托咪酯是否也會(huì)直接誘發(fā)人肝癌HepG2細(xì)胞的凋亡呢?我們提出了該研究的假設(shè)。本文設(shè)計(jì)不同濃度的依托咪酯是否具有直接誘導(dǎo)該細(xì)胞凋亡的作用,2.1.1 細(xì)胞凍存 取1mL的胰酶消化2min后,將對(duì)數(shù)生長(zhǎng)期的細(xì)胞從培養(yǎng)瓶表面貼壁沖洗下去,并懸浮于3mL的培養(yǎng)液中,用1000rpm離心5min,去上清液。將人肝癌HepG2細(xì)胞懸浮于1mL的凍存液中,調(diào)整細(xì)胞濃度至106/mL,每個(gè)凍存管中加入1mL的細(xì)胞凍存液,放置-20℃冰箱內(nèi)保存2h,然后置于-80℃冰箱過(guò)夜。

2.1.2 細(xì)胞復(fù)蘇 將凍存?zhèn)溆玫募?xì)胞,放入37℃的水浴箱內(nèi)融化復(fù)蘇。

2.1.3 細(xì)胞培養(yǎng) 將人肝癌HepG2細(xì)胞放置于37℃、5% CO2細(xì)胞培養(yǎng)箱中培養(yǎng),每3天傳代培養(yǎng)1次,細(xì)胞貼壁達(dá)到70%時(shí),以0.25%胰酶消化,使細(xì)胞全部都浸于溶液,倒置顯微鏡下觀察,細(xì)胞變圓后,但尚未漂起時(shí)加入5mL培養(yǎng)液終止消化,然后分到另兩個(gè)培養(yǎng)瓶中,加入新鮮培養(yǎng)基后再放置于37℃、5%CO2細(xì)胞培養(yǎng)箱中繼續(xù)培養(yǎng),取對(duì)數(shù)生長(zhǎng)期細(xì)胞。

2.1.4 細(xì)胞傳代 采用倒置顯微鏡觀察細(xì)胞貼壁生長(zhǎng)狀況,當(dāng)生長(zhǎng)至80%以上時(shí),用0.25%胰酶消化進(jìn)行傳代,并置于37℃、5%CO2細(xì)胞培養(yǎng)箱中繼續(xù)培養(yǎng)。

依托咪酯為臨床常用的一種麻醉劑,在靜脈麻醉過(guò)程中只需一次給藥就可以獲得很好的效果,但是依托咪酯所產(chǎn)生的毒性作用也是麻醉劑中獨(dú)有的,它可以促進(jìn)免疫細(xì)胞的凋亡。許多國(guó)內(nèi)學(xué)者研究依托咪酯誘導(dǎo)腫瘤細(xì)胞凋亡作用,從而分析依托咪酯的藥理學(xué)和臨床治療的意義。有研究表明[5-8],小劑量依托咪酯對(duì)人急性髓性細(xì)胞白血病細(xì)胞具有一定的抑制作用。

為探討不同濃度的依托咪酯是否具有直接誘導(dǎo)人肝癌HepG2細(xì)胞凋亡的作用而進(jìn)行該試驗(yàn)。采用小劑量和大劑量的依托咪酯做細(xì)胞凋亡分析,綜合相關(guān)文獻(xiàn),參照依托咪酯對(duì)外周血單個(gè)核細(xì)胞凋亡影響的試驗(yàn)方法及藥物劑量規(guī)格[9-12],觀察該細(xì)胞正常時(shí)細(xì)胞凋亡情況,以及分別給予小劑量0.5μg/mL,5μg/mL的依托咪酯、大劑量50μg/mL后12h、2h、48h依托咪酯時(shí)對(duì)人肝癌HepG2細(xì)胞凋亡的情況。實(shí)驗(yàn)結(jié)果證明依托咪酯誘導(dǎo)人肝癌細(xì)胞株HepG2細(xì)胞的凋亡具有一定的濃度-效應(yīng)與時(shí)間-效應(yīng)的關(guān)系,為依托咪酯開(kāi)辟臨床治療的新途徑。
